Abstract

Background: Cholangiocarcinoma (CCA) is a cancer of the biliary tract that is increasing in incidence. The Hippo pathway effector Yes-associated protein (YAP) has been identified by us and others as a driver for oncogenesis. YAP canonical regulatory pathways consist of serine kinases. These kinases are rarely altered in CCA but increased YAP activity is consistently observed. We have previously demonstrated that YAP tyrosine phosphorylation serves as an important, activating, pathway in CCA. We have also identified the YAP-interacting tyrosine phosphatase, SHP2, as an inhibitory modulator of this pathway and CCA growth. Herein, we explore an additional tyrosine phosphatase, protein tyrosine phosphatase 1B (PTP1B), and its role in regulating YAP activity and CCA cell growth.
