Abstract

Recently, formation of lipofuscin-like substances has been observed in a reaction mixture of rabbit erythrocyte ghost membranes with t-butylhydroperoxide (t-BOOH), which were found to be derived from linoleic acid hydroperoxide and phosphatidyl ethanolamine in the presence of haem, but not from malondialdehyde (MDA) and 4-hydroxynonenal (4-HN). Dietary antioxidants, in particular water-soluble antioxidants were found to inhibit formation of lipofuscin-like substances, although lipid-soluble antioxidants had no effect. However, sesaminol, a novel lipid-soluble antioxidative lignan isolated from sesame oil, was found to prevent effectively both lipid peroxidation induced by carbon tetrachloride (CCl4) and excretion of 8-hydroxy-2’-deoxyguanosine (8-OH-dG) in urine determined by monoclonal antibody method using in vivo system. These effects were also observed in the novel lignan glucosides isolated from sesame seed. Detailed examination has been carried out to find the contribution of phenolic and β-diketone groups for the antioxidative mechanisms.
