Abstract

Objective To investigate the protective effects of Zuogui Pill on oxidative stress in rats with chronic cerebral ischemia.Methods All rats were divided into control,sham operated,chronic cerebral ischemia and Zuogui Pill treatment group.Rats in later two groups were subjected to permanent bilateral common carotid artery occlusion.At 3 weeks after operation Zuogui Pill was begin to administered orally by gavage to Zuogui Pill treatment group rats at doses of 4.0 g/kg per day.finally 8 rats were in each group.At 6 weeks after operation All rats were tested by water-maze.Reactive oxygen species (ROS) production and respiratory function in rat brain mitochondria was monitored.Results Compared with chronic cerebral ischemia group,rats in Zuogui Pill treatment group had shorter escape latency [(36.43 ± 6.51)vs.(43.46 ±4.28) s,P < 0.05],decreased ROS level (120.52 ± 10.80 vs.137.46 ± 15.28,P <0.05),lower levels of respiration control rate and oxidative phosphorylation parameters (P < 0.05).Conclusion Zuogui Pill could improve mitochondrial respiratory function and decrease elevated level of ROS,which may be the mechanism of its protection against Oxidative Stress in Rats with Chronic Cerebral Ischemia.
