Abstract

Objective To observe the protective effect of Lidan Xiaohuang decoction on hepatic ischemia reperfusion injury in rats. Methods 225 SD rats were randomly divided into sham-operated group (Sham group), ischemia reperfusion injury (HIRI) control group (control group) and HIRI traditional Chinese medicine group (TCM group). Each group was divided into three subgroups at 60, 180 and 360 min after reperfusion. Rats in TCM group were given Chinese medicine gavage before operation. Control group and Sham group were given the same volume of normal saline. In Sham group, only open abdominal operation was performed, and TCM group and control group were treated with HIRI. A rat HIRI model was established and blood was collected from the inferior vena cava at 60, 180 and 360 min after ischemia and reperfusion in the liver. Alanine aminotransferase (ALT), aspartate aminotransferase (AST) and lactate dehydrogenase (LDH) levels were measured, and fresh liver specimens were taken at the same time for measuring MDA content and GSH-PX activity, and histopathological observations were performed. Results After 60, 180 and 360 min of HIR in rats, the ALT, AST and LDH in TCM group were (74.52±8.95), (89.48±16.87), (118.95±18.47), (225.94±49.76), (276.36±78.95), (329.85±79.59), (1 467.44±295.46), (2 860.59±347.32), (3 020.22±357.83) U/L respectively. The MDA content and the activities of GSH-PX were respectively (3.94±0.86), (7.33±3.12), (12.89±6.48) nmol/mgprot and (1 866.34±364.49), (1 299.05±269.49), (780.77±185.03) U/mgprot; The ALT, AST and LDH levels in the control group were respectively (99.24±12.71), (182.37±21.62), (298.13±36.26), (356.87±57.36), (529.31±79.39), (875.69±83.61), (1 937.59±279.55), (4 207.43±654.87), (4 672.66±539.11) U/L. And the MDA content and the activity of GSH-PX were (6.76±1.56), (14.20±4.87), (25.82±9.86) nmol/mgprot and (1 689.33±339.73), (792.19±241.66), (429.80±126.07) U/mgprotrespectively. As compared with the control group, the levels of ALT, AST, LDH and MDA contents in the TCM group were significantly reduced, and the GSH-PX activity was slightly decreased (P=0.000); The changes of ALT, AST and LDH levels, MDA content and GSH-PX activity in the Sham group were smaller (P=0.000). Pathological observation showed that TCM group and Control group also had a significant difference. Conclusion Lidan Xiaohuang decoction can protect against HIRI in rats, and the protective mechanism is related to its antioxidant mechanism.
