Abstract

We studied the effect of prostaglandin E 1 (PGE 1) and prostacyclin (PGI 2) infusion on thromboxane A 2 (TxA 2) production in endotoxin-induced lung injury by using a chronic lung lymph fistula model in sheep. Seven sheep received alternately Escherichia coli endotoxin and endotoxin plus PGE 1 (1 μg/kg/min). Six sheep received PGI 2 (0.2 μg/kg/min) instead of PGE 1. The infusion of both PGE 1 and PGI 2: (1) decreased TxA 2 production (as measured by RIA of its metabolite TxB 2); (2) decreased pulmonary hypertension; and (3) decreased interstitial edema. The effect of PGE 1 and PGI 2 infusion on TxA 2 production may be independent of effects on the reaction of platelets and leukocytes.
