UENPS.321 Prophylactic methods of providing fluorosis in rats' offspring skeletal system used in perinatal period Elzbieta Kolodziejczak⁎, Jerzy Szczapa, Iwona Inkielewicz, Wojciech Czarnowski, Krzysztof Preis Medical University, Gdansk, Poland Medical University, Poznan, Poland Background and aim Fluoride (F–) is a natural component of biosphere. It appears in soil, air and water. In the human organism approximately 99% of the fluoride is associated with calcified tissues. Excessive intake of fluoride may lead to pathological changes in bones — skeletal fluorosis. The purpose of the study was to find a prophylactic method to provide bone fluorosis in rats' offspring when their mothers received different solutions of Fconcentration. Materials and methods We used 60 pregnant female (first generation) Winstar rats and divided them into three groups according to F– concentration in drinking water: 0.3 ppm (control), 20 ppm, 80 ppm. Each group of pregnant rats was fed different diets: XX control and enriched in: Calcium — CX, Vitamin D3 — XD, Calcium and Vitamin D3 — CD. Male rats' offspring (second generation) received F–, Calcium and Vitamin D3 during pregnancy through placenta and by breastfeeding with mothers milk. Rats were put into metabolic chambers and samples of urine were collected. Animals were euthanized after stopping weanling. The femoral bones were extracted. The fluoride concentration in bones was assessed. The bone maximal bending force F max (kN) was examined and bone bending resistance Rg (MPa) was counted according to the formula. The fluoride concentration in urine and bones was measured by a fluoride ion specific electrode and an Ag/AgCl refence electrode with double jacket. Calculations were based on a response factor from a standard curve prepared daily. Results Bone F max (kN) in rats' offspring's increased for 23% in CX diet and 62% in CD (20 ppm F–). In the group 80 ppm Fthe increase was for 10% CX and 33% CD. The differences were found statistically significant for the CD diet in both groups of F– concentration. The increase of bone Rg (MPa) in second generation was statistically significant only for 80 ppm F– it rose by 19% XD, 23% XC and 27% CD. Fluoride bone concentration (mgF/g) decreased for 7% XD, 12% CX, 25% CD (20 ppm F–) and 25% XD, 31% CX, 35% CD (80 ppm F–). The differences were found statistically significant. Conclusions High Fluoride doses administered to rats mothers during prenatal period lead to its accumulation in bone system and diminish mechanical bone parameters of their offspring. Higher intake of Calcium and Vitamin D3 by pregnant and breastfeed mothers improve the resistance of offspring's bones. doi:10.1016/j.earlhumdev.2008.09.337 Abstract UENPS.322 In vitro fertilisation(IVF) births in BitolaUENPS.322 In vitro fertilisation(IVF) births in Bitola Meri Dzarlieva⁎ Clinical hospital, Bitola, Macedonia Background and aim Infertility is a big challenge nowadays all over the world. This introduced the need for IVF in Bitola since the second half of 2004. Materials and methods Material have been taken from the newborns history/list from our department. Results Using this method, a total of 14 women gave birth to 14 newborns. In 2005, a total of 39 newborns were born, including 8 twins and 2 triplets. In 2006, 5 twins and 2 triplets were born amongst 44 newborns. In 2007, a total of 69 newborns were born, of which 5 were twins and 2 triplets. In the first third of this year there have been 31 births, including 2 twins. Women from all over Macedonia are treated in this private health organization but they often deliver in their local hospitals. Conclusions Since the introduction of this method in Bitola, there have been a total of 197 newborns, which include 40 twins and 18 triplets.
