Abstract
Objective To study the inhibitory effect of four kinds of cryoprotectants, including glycerin, dimethyl sulfoxide (DMSO), ethylene glycol and propylene glycol, on K562 cells (Chronic myeloid leukemia cell line). Methods The effects of the four cryoprotectants on the proliferation activity of K562 cells in vitro were investigated by the methods of trypan blue exclusion, MTT assay and flow cytometry. Results At the final cryoprotectant concentration of 3%, cell viability were significantly reduced compared with control groups. But K562 cells maintained in 1% glycerin or DMSO solution kept their viability well, and the former was safer than the latter. 0. 5% Propylene glycol could inhibit the proliferation of K562 cells. Conclusion Residual cryoprotectants may inhibit cell viability at low concentration, and glycerin was the most optimal to cells among these four cryoprotectants.
