Abstract

目的 研究纳米氧化硅诱导巨噬细胞生成细胞因子在成纤维细胞增殖和胶原合成中的作用.方法 31.25、125.00、500.00 μg/ml纳米氧化硅及500.00 μg/ml微米SiO2对巨噬细胞系RAW264.7染毒24 h,用酶联免疫(ELISA)法测定培养上清液中肿瘤坏死因子α(TNF-α)、转化生长因子β1(TGF-β1)及白细胞介素-1β(IL-1β)的表达;取上述浓度尘粒染毒RAW264.7细胞培养上清液与中国全鼠肺成纤维细胞(CHL)共同孵育后,分别用噻唑蓝(MTT)进行比色分析法,用消化法测定CHL细胞增殖率及羟脯氨酸生成量.结果 125.00、500.00μg/ml纳米氧化硅组及500μg/ml微米SiO2组TNF-α[(19.97±0.76、22.76±2.69、24.09±1.44)pg/ml]、TGF-β1[(53.07±3.88、58.78±5.48、59.75±7.93)pg/ml]及IL-1β(11.76±0.82、12.71±0.30、15.08±2.18)pg/ml]含量明显高于阴性对照组,差异有统计学意义(P<0.05);125.00、500.00μg/ml纳米氧化硅组及500μg/ml微米SiO2组诱导CHL细胞增殖率及羟脯氨酸含量较阴性对照组增加,差异有统计学意义(P<0.05);500.00μg/ml纳米氧化硅组CHL细胞中羟脯氨酸含量高于500 μg/mg微米SiO2处理组,差异有统计学意义(P<0.05).随纳米氧化硅染毒浓度的增加,细胞因子的含量、CHL细胞增殖率及羟脯氨酸生成量逐渐升高.结论 纳米氧化硅在介导巨噬细胞释放大量细胞因子、诱导肺成纤维细胞增生和胶原纤维生成增多方面,具有与微米SiO2同样的作用。
