Abstract

At present, the use of antibiotics and growth hormones for livestock has been prohibited by the regulations. Probiotics are one of the additives that are safe and permitted by regulations, and also can replace the use of antibiotics. Probiotics are live microorganisms used in livestock to improve microbial balance and have positive effects on host animals. The purpose of this study was to optimize the production of probiotic powder media by using Central Composite Design-Response Surface Methodology (CCD-RSM). The RSM-CCd was prepared using four variables, namely starch, CaCO3, Corn Gluten Meal (CGM), and zeolite, with five-level combinations. The drying process of probiotic powder used a vacuum oven at 45°C for 24 hours. The vacuum process was carried out using a vacuum pump for three hours. L. plantarum TSD-10 survival amount was measured by the total plate calculation method. Statistical analysis showed that the viability of probiotics was significantly affected by CaCO3, CGM, and zeolite. According to the model developed, the amount of optimum L. plantarum TSD-10 surviving was estimated can reach around 100.01%, with the content of starch, CaCO3, CGM, zeolite are 30%, 45%, 30%, 60% respectively.
