Abstract

Messenger RNA was prepared from first instars of the common cattle grub, Hypoderma lineatum (Villers). Larvae were excised from esophagi of cattle and frozen in liquid nitrogen. Messenger RNA was extracted by grinding the frozen tissue with hot phenol, partitioning the nucleic acids into an aqueous phase, precipitation with ethanol, and chromatography on oligo(dT)-cellulose. Poly(A)-RNA extracted by this method retained biological activity as demonstrated by in vitro translation using rabbit reticulocyte lysate and [35S]-Met. H. lineatum proteins produced by in vitro translation were analyzed by dodecylsulfate-polyacrylamide gel electrophoresis and autoradiography. Immunoaffinity isolation of specific H. lineatum proteins demonstrated translational fidelity of the mRNA.
