Abstract

A method is presented for the isolation of nonhistone chromosomal proteins from chromatin of human diploid WI-38 fibroblasts which involves dissociation of total chromosomal protein from DNA followed by separation into a histone and a nonhistone fraction. Loss of nonhistone protein is minimized at each step, and the only substantial loss, during chromatin dissociation, is shown to be random. Finally, the nonhistone chromosomal protein fraction is separated into five major fractions by column chromatography on QAE Sephadex.
