Abstract
This chapter discusses the pharmacological models to study opiate dependence. For measuring analgetic activity, the battery of tests proved to be useful, such as hot plate test, writhing test , tail flick test, and algolytic test. The analgetic effect of morphine, hydromorphone, and oxymorphone was compared with two newly synthetized opiate analgetics: azidomorphine and 14-hydroxyazidomorphine. It is evident from the data obtained from the study that animal tests allow the prediction of expectable analgetic potencies of new compounds in man. The new derivatives, azidomorphine and 14-hydroxyazidomorphine, which were more potent than hydromorphone and oxymorphone in all the tests used, also proved to be significantly more effective in clinical trials.
