Abstract

The aim of this study was to investigate the effects of combined exercise program during weeks on the body composition, physical fitness and blood biochemical parameters according to the Pro12Ala polymorphism in the PPAR-γ2 gene in obese middle-aged women. A total of 34 participants were recruited in this study, and they performed combined exercise during weeks. Polymerase chain reaction(PCR)-and BstUI restriction fragment length polymorphism(RFLP) analysis was performed to analyze the Pro12Ala polymorphism in the PPAR-γ2 gene. Combined exercise program indicated the beneficial effects on the body composition, physical fitness and blood lipid profile and glucose metabolism of the subjects(p.05). The genotype distribution of the Pro12Ala polymorphism in the PPAR-γ2 gene were 85.3% for Pro12Pro genotype, and 14.7% for Pro12Ala genotype, respectively, and observed genotype frequencies were in Hardy-Weinberg equilibrium. However, this polymorphism was not significantly associated with any clinical parameters studied, and the significant differences between two genotypes were not observed in the changes of any metabolic parameters following combined exercise. Although, combined exercise program indicated the health-promoting effects on the mitigation of lifestyle-related disease such as obesity and non-insulin dependent diabetes mellitus(NIDDM) for the subjects, the Pro12Ala polymorphism in the PPAR-γ2 gene may not significantly contribute to the inter-individual differences on the clinical effects by combined exercise program. Further studies using larger smaple size and other candidate genes will be needed.
