Abstract

The modern technological approach in the research area of polymer therapeutics has profound impact on the development of the polymer–drug conjugates. The biocompatible and biodegradable polymer used for different drug conjugates have the advantages of prolong blood circulation, improved pharmacokinetics and better tumor tissue localization by passive targeting than the native drugs. Polymers such as N-(2-hydroxypropyl methacrylamide) (HPMA) copolymers, poly-l-glutamic acid (PG), and poly(ethylene glycol) (PEG) are used for conjugation of potent therapeutic drugs for enhanced clinical efficacy. Further, the therapeutic efficacy is improved by conjugating appropriate targeting moieties to the polymer–drug conjugates, helps in delivering the drug to the appropriate site of action. In this chapter, the rational design of polymer–drug conjugates chemistry and their potential applications towards therapeutics are discussed.
