Publisher Summary This chapter discusses the current advances with respect to the genes expressed in pollen that have been isolated, the identification of these genes with respect to function, and the makeup of promoters that direct the pollen-specific expression of the genes under their regulation. Transcription of genes occurs from the haploid genome following meiosis of microsporocytes and during the subsequent differentiation of the microspores into mature male gametophytes. A large number of different genes are transcribed during postmeiosis microspore and pollen development. Several cDNA clones corresponding to genes expressed in the developing male gametophyte are isolated, sequenced, and a few tentatively identified on the basis of function. The pollen-specific cDNA clone from maize, Zm13 , is a full-length copy of the mRNA, which is 929 nucleotides long. Zm13 codes for a predicted polypeptide of 170 amino acid residues with a molecular mass of 18.3 kDa. The hydropathy profile of the polypeptide suggests a possible signal sequence at the amino terminus.
