Abstract

A polarographic method is described for the qualitative and quantitative determination of 3-keto sugars. For highest sensitivity, the polarograms are recorded at pH 11 to 11.5 in 0.1 M LiCl as indifferent electrolyte. Under these conditions, as little as 3 × 10 −6 M of a 3-ulose can easily be identified. For all the different 3-uloses measured, the characteristic feature, E 1 2 , ranges between −1.50 and −1.57 V vs the saturated calomel electrode. In addition, recordings at neutral pH indicate to what extent the carbonyl group of a 3-ulose is hydrated. The method was applied to establish the substrate specificity of the 3-ulose forming enzyme hexopyranoside: cytochrome c oxidoreductase.
