Abstract
The feasibility of using progesterone and oestrone sulphate (E 1 S) levels for oestrus confirmation and early pregnancy diagnosis was studied in 15 buffaloes. Profiles of progesterone, oestrone and E 1 S were determined in 10 buffaloes from day 0 to day 205 after AI. Basal levels of E 1 S were below 100 pg/ml. Five pregnant animals showed slight increases in E 1 S at day 51 (104·46 ± 64·04 pg/ml) and day 86 (102·10 ± 51·32 pg/ml) and gradually increased after day 100 to reach 1260·80 ± 259·17 pg/ml at day 205. Five animals which aborted showed levels below 100 pg/ml from days 0 to 205 after AI. At day 20 after AI, E 1 S levels in pregnant animals were significantly higher than in the non-pregnant and aborting animals: 138·03 ± 67·80, 51·04 ± 19·12 and 56·20 ± 25·73 pg/ml, respectively. Radioimmunoassay of progesterone without extraction is described and validated. Application of progesterone and E 1 S determinations for oestrus confirmation and early pregnancy diagnosis in buffalo in conjunction with an AI service are discussed.
