Abstract

Summary Leaf mesophyll protoplasts of Broussonetia kazinoki Sieb. were enzymatically isolated from in vitro grown shoot cultures. The protoplasts showed first divisions between 10-20 days in culture and formed colonies within 4 weeks in modified Murashige and Skoog (MS) medium. Reduction of ammonium concentration and use of glucose as a carbon source were essential for sustained cell divisions and colony formation. Shoot buds were produced from protoplast-derived callus on modified MS medium containing 0.1 mg · 1 −1 NAA and 1 mg · 1 −1 BA.
