Abstract

The placental transfer of hexachlorophene (HCP) in albino rats was studied using material labeled with 14C in the methylene bridge. Pregnant rats were treated orally (10 mg/kg) and dermally (8 mg/kg) with a single dose of the material in an aqueous carboxymethyl cellulose suspension. Animals were killed at various times posttreatment and the concentration of HCP (based on 14C activity) in maternal blood, placental tissue, amniotic fluid, and the fetus was determined. Measurable amounts of HCP were detected in all tissues as early as 0.5 h following oral treatment. Placental transfer of HCP was demonstrated following both routes of administration. The transfer of HCP into milk was measured by dosing lactating rats and determining the HCP concentration of the milk coagulate of suckling rats. HCP was detected in the milk taken 1 hr or more after maternal treatment.
