Abstract

<p>Phycocyanin is a pigment-protein complex from the light-harvesting phycobiliprotein family which is<br />often found in cyanobacteria. The product phycocyanin produced by phanizomenon flos-aquae and Spirulina<br />sp. The aim of this study were to determine the best solvents purification phycocanin from Spirulina platensis<br />in three solvents, phosphate buffer, water and aceton ammonium sulphate and to evaluate the antimalarial<br />activity in vitro of phycocyanin in the best solvent extraction from S. platensis. The method of this study was<br />using in-vitro antimalarial method. The result showed C- phycocyanin (C-PC), yield, and protein contents<br />of phycocyanin were 8 mg/mL, 20.22%, 1.88% extracted and purified by phosphate buffer, 6.63 mg/mL,<br />16.58 %, 3.51% extracted and purified by water, 2.86 mg/mL, 7.15%, 8.4% extracted and purified by acetone<br />ammonium sulphate respectively. Phosphate buffer was the best solvent of phycocyanin extraction from S.<br />platensis. Antimalarial activity in vitro of phycocyanin in hosphate buffer against Plasmodium falciparum<br />strains 3D7 with IC50 was 158,489 μg/mL. The possible mechanism might be relied on the destruction of<br />polymerization of Haemozoin by binding of C-PC with ferriprotoporphyrin-IX at the water surface of the<br />plasma membrane.</p>

BAHAN DAN METODE Bahan dan Alat

Bahan-bahan yang digunakan dalam penelitian ini meliputi kapsul Spirulina komersial, air tawar, air laut, inokulum Spirulina platensis diperoleh dari Balai Besar Pengembangan Budidaya Air Payau (BBPBAP) Jepara-Jawa Tengah, media Walne, etanol, bufer fosfat, akuades, aseton, dan amonium sulfat, BSA, Folin-ciocalteu-fenol, Cu-alkali, HCl 2N, artemisinin, P. falciparum 3D7 (cloroquine sensitive) yang diperoleh dari Lembaga Biologi Molekuler Eijkman, RPMI 1640 (Sigma-Alorich), HEPES (Sigma), NaHCO3 (Sigma®), disinfektan (baycline), Na2HPO4, antibiotik gentamisin injeksi (Sigma® G1397), NaCl 3,5 %, sorbitol 5%, serum darah, sel darah merah, antikoagulan sitrat fosfat dektrosa (CPD), giemsa (Merck KGaA), akuabidestilata, nitrogen cair. Kultivasi S. platensis mengacu pada Diharmi et al (2001) dengan cara S. platensis dikultivasi menggunakan media Walne, suhu 25oC, intensitas cahaya 3000 lux perbandingan terang: gelap 16:8 (jam), salinitas air laut 15 ppt dan bibit yang digunakan 20% dari volume kultur. Ekstraksi fikosianin dilakukan dengan cara serbuk Spirulina diekstrak menggunakan tiga jenis pelarut yang berbeda yaitu bufer fosfat, air, dan aseton. Ekstraksi fikosianin menggunakan bufer fosfat dan air mengacu pada Silveira et al (2007) yang dilakukan dengan cara melarutkan serbuk Spirulina 0,04 g menggunakan 1 mL bufer sodium fosfat 10 mM pH 7,0 (NaOH-KH2PO4) atau air. Ekstraksi menggunakan aseton mengacu pada Pankaj et al (2010) yang dilakukan dengan cara serbuk Spirulina dilarutkan menggunakan aseton 80% kemudian diinkubasi selama 24 jam dengan suhu 4oC. Pelet hasil sentrifugasi dilarutkan kedalam 50 mM bufer fosfat pH 7,0 kemudian suspensi disentrifugasi pada 3.500 rpm, suhu 4oC, selama 5 menit.
