Abstract
Here we report a photosensitive supramolecular hydrogel formed by MeO-Tet-GA. MeO-Tet-GA can self-assemble to form supramolecular hydrogel in neutral buffer. The formed hydrogel has good mechanical properties, and the dynamic energy storage modulus G ′ is above 500 Pa. Based on the intermolecular photo-click reaction between biaryltetrazole and the terminal alkene, we could further modify the hydrogel with hydrophilic sodium 4-vinylbenzenesulfonate to disturb the hydrogel and induce the degradation of hydrogel. We also tried to use this supramolecular hydrogel for three-dimensional culture of mouse breast cancer 4T1 cells. We found that MeO-Tet-GA has excellent biocompatibility and did not cause significant 4T1 cell death after three days of culture. Overall, this work demonstrates that supramolecular hydrogel formed by MeO-Tet-GA holds great promise in long-term three-dimensional cell culture and in studying cell behaviors through photo-modulation of cell microenvironment.
