Photochemical cleavage of oligonucleotides from solid phase supports

Abstract

5′-O-Dimethoxytrityl-3′-O-succinatothymidine is covalently linked to long chain alkyl amine controlled pore glass support (LCAA-CPG) via an o-nitrobenzyl group. The modified solid phase synthesis support is compatible with standard automated phosphoramidite based oligonucleotide synthesis. Cleavage from the solid support is achieved via CuSO 4 filtered photolysis. Isolated yields are comparable with those obtained from other oligonucleotide supports.
