Abstracts S82 Figure 1 Crkl SH2 domain complexes in vivo with tyrosine-phos-phorylated Hef1/Cas-L in lymphomas of P190 BCR/ABL transgenicmice. (a) Crkl is associated in vivo with Hef1 and P190 Bcr/Abl. Pre-cleared lymphoma extract was immunoprecipitated with Crkl antisera(lane 2) or monoclonal anti-phosphotyrosine antibodies (lane 3), sep-arated on 7.5% SDS-polyacrylamide gels and transferred to poly-vinylidene membranes. As control, total P190 lymphoma lysate wasloaded (lane 1). Duplicate blots were reacted with antibodies againstphosphotyrosine (upper panel) or Cas/Hef1 (lower panel). (b) Far-Western blot analysis of P190-Bcr/Abl lymphoma immunoprecipit-ates. Lane 1 contains total P190 Bcr/Abl leukemia/lymphoma lysate.Lysate proteins were immunoprecipitated with antibodies directedagainst Cas/Hef1 (lane 2) or phosphotyrosine (lane 3). Western blotwas incubated with GST-Crkl SH2 and bound protein was detectedwith anti-GST antibodies. Molecular weight markers are indicated atthe left, locations of the detected proteins are shown to the right.
