Abstract

Pharmacokinetic studies of cefazolin (CFZ) were conducted after single dose (30 mg/kg, IV) in afebrile goats and febrile goats with and without probenecid (1 g, IV). The zero time plasma concentration ( C p 0) in febrile goats with probenecid(PB) was higher ( p < 0.05) compared with the febrile goats-without PB. The PB administration prolonged the time of the therapeutic plasma concentration (Cp ther) in the febrile group with PB as compared to without PB. Concurrent administration of PB with CFZ decreased ( p < 0.05) the values of total body clearance (cl B), renal clearance (cl R) and hepatic clearance (cl H) of CFZ in febrile goats. Volume of distribution is usually expected to change when a febrile state is induced. Lack of such an effect in this study could be due to high individual variation. PB could block tubular secretion of CFZ and prolong the Cp ther in febrile goats when given concurrently with CFZ. It is concluded that probenecid may be given IV with CFZ especially when higher plasma concentrations of CFZ are required for longer duration in febrile goats.
