Abstract
Abstract Isolated bovine thyroid cells, incubated at 37 ° C, take up latex beads as shown by the UV (ultraviolet) absorption curve of dioxane cell extracts and by electron microscopy. Thyrotropin (TSH), prostaglandin E 1 (PGE 1 ), and dibutyryl cyclic AMP (DBcAAAP), each of which has been shown to stimulate the endocytotic process in thyroid, augment phagocytosis of latex beads in isolated thyroid cells in a dose-related manner. The membrane stabilizer, chlorpromazine (CPZ), at low concentrations inhibits TSH, PGE 1 , and DBcAMP effects on bead phagocytosis, while at higher CPZ concentrations, polystyrene uptake by cells is increased, suggesting membrane destabilization. These findings suggest that phagocytosis of latex beads by isolated thyroid follicular cells is a model for the phagocytosis of colloid involved in thyroid hormone secretion. The technique provides a convenient system for direct investigation of the thyroidal phagocytotic process in vitro.
