However, very little is known about their possible role in bladder cancer development. Here, we evaluated the role of MDSC in bladder cancer. METHODS Flow cytometry was performed on peripheral blood of 113 patients with bladder cancer and 20 healthy controls with a median follow-up of 43 months for overall survival (OS). We also assessed the expression of MDSC-inducing cytokines. RESULTS For the ?rst time, we characterized CD33-CD11b+HLA-DR- cells sorted from the peripheral blood of BC patients. CD33-CD11b+HLA-DR- cells suppressed autologous, antigen-independent T cell proliferation. The frequency of MDSC correlated with tumor grade and stage. We also showed that increased levels of circulating MDSCs correlate with reduced overall survival. Increases in the levels of pro-MDSC cytokines were observed for bladder cancer patients versus controls, and the pro-MDSC cytokine IL-6 was decreased in those patients, down-regulation of IL6 is sufficient to induce MDSC formation in vitro.
