Abstract

This paper set up a reliable, time-saving and simple technique, PCR-LOH, for assay of chromosome 1p deletion in neuroblastoma. The authors applied polymerase chain reaction (PCR) to 8 cases of genotype neuroblastoma DNA at DIS 80 locus containing a variable number of tandem repeats which is located on the distal part of short arm of chromosome 1. Agarose gel electrophoresis and ethidium bromide staining of the amplified products could make a simple determination of 6 cases with loss of heterozigosity (LOH) of the short arm of chromosome 1. This technique may be completed in one work day and makes it possible as a routine detection for chromosome 1p deletion in neuroblastoma. Key words: PCR-LOH; Chromosome 1p deletion; Neuroblastoma
