Abstract
Aerosolized delivery of drugs to the lungs is the most preferred route of drug administration for the treatment of respiratory and other ailments. It is employed for the pulmonary delivery of a wide range of therapeutic agents, right from small molecules to biologics. Particle engineering to generate, design, and control particles with desired properties is vital in the formulation and manufacturing of micro- and nano-sized drugs for consistent and efficient drug delivery to the lungs. In this chapter, we describe the significant particle properties and key particle engineering strategies for the formulation development and manufacturing aspects of pulmonary drug delivery systems. Several modern and advanced techniques are discussed that have shown an exciting potential to be developed into a platform technology providing a quick, cost-effective, safe, and sustainable solution for particle engineering aimed at pulmonary drug delivery applications. Besides this, an outlook towards promising opportunities for new modalities in the pulmonary drug delivery landscape is briefed.
