Abstract

Lipoxygenase activities in ungerminated and germinating barley grains were found to be associated exclusively with the embryos. A lipoxygenase was extracted from ungerminated embryos and partially purified by fractional precipitation with ammonium sulfate and gel-filtration. Both the crude extracts and the purified preparation appeared to contain only a fatty acid type lipoxygenase which mainly converted linolele acid to 9-hydroperoxy, trans-10, cis-12-octadecadienoic acid. The purified enzyme was inhibited by its own products, hydroperoxides, but not by 1 mM cyanide, EDTA, Hg2+ or Cu2+.
