Abstract

Background and Aims: Paraoxonase 1 (PON1) is an antiatherogenic high density lipoprotein (HDL)-associated lactonase. Previous findings revealed that PON1 knockout (PON1KO) mice have low blood pressure, which is inversely correlated with their levels of 5,6-epoxyeicosatrienoic acid (5,6-EET), a cytochrome P450 (CYP)-derived arachidonic acid (AA) metabolite. Our previous studies revealed that 5,6-EET is unstable, transforming to the δ-lactone isomer 5,6-δ-DHTL, which is a potential endothelium-derived hyperpolarizing factor (EDHF) that mediates vasodilation, and it is also a potential substrate for PON1.
