P017 Automation of HLA-typing using next generation sequencing with Omixon’s holotype HLA X2 kit and beckman-coulter’s biomek liquid handling system

Abstract

Holotype HLA is an amplification and library preparation kit from Omixon designed to provide high resolution HLA typing using Next Generation Sequencing (NGS). While Holotype HLA is designed with the technician in mind, consistency and throughput can be increased using Biomek liquid handling systems. Using Coriell DNA samples that have been typed and validated manually, four separate runs with variable sample volumes will be processed on Biomek liquid handlers from Beckman–Coulter. Pre-PCR and library preparation will be done separately, and results from the automated runs will be compared with manually run Holotype HLA kits. Concordance between manually run Holotype HLA kits and automated runs on the Biomek liquid handling systems are comparable, but the Biomek liquid handling system has a much lower hands-on time allowing for increased throughput. Automating the Holotype HLA kit on Biomek liquid handling systems generates accurate NGS typing results and decreases hands-on time. R. Pollok: Employee; Company/Organization; Omixon.
