Abstract
The chemical oxidation of tyrosine with potassium nitrosodisulphonate (Fremy salt) was found to follow step by step the enzymatic pathways, i.e. —NH 2 terminal tyrosine is oxidized to 2-carboxy-5,6-dihydroxy-indole through the intermediate of “dopachrome”, while tyrosine in the middle of a peptide chain, or with —COOH terminal follows the “dopaquinone” pattern yielding 3,4-dihydroxy-phenylalanine derivatives. The spectroscopic data have been analysed and the method is proposed as a sensitive assay replacing the enzymatic method.
