Abstract

Nonprotein sulfhydryl groups (glutathione, cysteine, coenzyme A) were rapidly oxidized by oxygen, the rate of oxidation being proportional to the oxygen pressure. The -SH groups of yeast alcohol dehydrogenase were also oxidized by oxygen at high pressure, and the enzyme activity was inhibited. The oxidation of unsaturated fatty acids, of hemoglobin, and of dihydrodiphosphopyridine nucleotide was not affected by oxygen at high pressure. It is postulated that the harmful effects of oxygen are due mainly to the oxidation of the -SH groups which are necessary for the cell activities.
