Abstract

Rabbits were pretreated 0.5 to 48 hr with alpha-methyl meta-tyrosine (α-MMT) prior to challenge with a single toxic dose of ouabain. The pretreatment of rabbits with α-MMT, in many cases, prolonged the time to ventricular rhythm disturbances induced by the single intravenous dose of ouabain. The protection afforded by such pretreatment appeared to be biphasic. The early protection, after only 1 2 hr of pretreatment, against ventricular extrasystole seemed to be related to a fall in blood pressure. However, the prolongation of the time to fibrillation and death was not totally related to blood pressure changes. That protection observed after longer pretreatment times was in no way related to blood pressure changes. The results obtained, in the present study, suggest that the adrenergic nervous system may be of importance in the development of ventricular arrhythmia in certain instances, but certainly does not seem to be the only mechanism involved in the development of ouabain-induced arrhythmia.
