Abstract

The use of inter-simple sequence repeats (ISSR) has been reported as being more useful for assessing genetic diversity for plant populations and inferring genetic relationships among closely related cultivars compared to standard DNA sequencing methods. In the present study, an optimal ISSR-PCR system was established for selected woody bamboos of economic importance. This was done by testing several PCR parameters, including annealing temperature, MgCl 2 concentration (1.5-2.5 mM), and primer concentration (1.5-2.5 μM). Three anchored primers were used. Gel electrophoresis was run using 1.8 and 2.0% agarose gel. Reproducibility of this method was tested by repeating the procedures under optimal conditions.
