Abstract

Cellulases are key enzymes in the biomass saccharification process. Basidiomycetes are capable of producing the entire complex of cellulases necessary for complete hydrolysis of the substrate. In research used a culture of wood-decay fungus I. lacteus Il-11 capable of active cellulose hydrolysis. The results showed that the citrate buffer system allowed stabilization of cellulases action. With increasing enzyme concentration, the amount of product increases but the rate of sugar formation decreases by 1.3–1.5 times with time (96–120 h). The value of the Michaelis-Menten constant is 0.063 M and the maximum reaction rate is 1.90 μM/min. The optimum concentration of cellulose for saccharification by I. lacteus enzyme complex is in the range of 4–7%. Glycine buffer promotes desorption of the I. lacteus cellulase complex from the substrate. Process flowsheet for the production and increase yield of I. lacteus cellulases is proposed.
