Abstract
A novel 20% concentration cross-linked agarose beads was prepared and successfully applied in the separation and purification of resveratrol and polydatin from crude root extracts of Polygonum cuspidatum. The optimal separation conditions were carried out with 20% acetic acid and 20% ethanol mixed solution (V/V) at a flow rate of 1.0 mL/min and a sample load of 16 mg crude extract (corresponding to a load of 1.0 mg/mL beads). The purities of the collected resveratrol and polydatin were more than 98.0% and the corresponding yields were 93.3% and 92.6%, respectively. The obtained 20% agarose beads were high chemical stability and load capacity, a low column back pressure, which could provide an alternative method for the separation and purification of active polyphenols that are frequently found in medicinal plants.
