Abstract

Isolated rat-liver nuclei were incubated in vitro in the presence of cortisol. The RNA polymerase prepared as “aggregate enzyme” according to Weiss, shows a significant increase in activity after 1 min of incubation of isolated nuclei with cortisol. The maximum of enzyme activation is reached after 10 min. Testosterone also caused an increase of RNA polymerase activity after 10 min of incubation of isolated rat-liver nuclei. Other steroids tested, hormonally active or inactive, showed no effect. The effect of cortisol on RNA polymerase activity is in good agreement with the increase in template activity of nuclear RNA, caused 15 min after incubation of isolated nuclei with this steroid.
