Abstract
ON THE ACTIVATION OF FATTY ACID OXIDATION
Highlights
University (Received for publication, August 27,1945). It has been previously reported from this laboratory that adenosine polyphosphates or compounds capable of phosphorylating adenylic acid are required to activate the oxidation of saturated fatty acids in crude rat liver homogenates [1]
Of activating fatty acid oxidation, since Ochoa has shown that Dhe reaction, a-ketoglutarate + 40, -+ succinat’e + CO, + HzO, is capable of vigorous esterification of inorganic phosphate [12]. ,4 direct trial of adenosine triphosphate (ATP), subst’ituted for adenylic acid + a-ketoglut,aret’e, was made, all other factors remaining constant
The slight activation by adenosine diphosphate (ADP) was possibly due to traces of ATP in the ADP
Summary
It has been previously reported from this laboratory that adenosine polyphosphates or compounds capable of phosphorylating adenylic acid are required to activate the oxidation of saturated fatty acids in crude rat liver homogenates [1]. In this paper the activation of fatty acid oxidation in prepared rat liver enzyme suspensionsmill be described These preparations have little or no endogenous oxidative activity and are much better adapted for the study of the activation than crude homogenates or the preparations of Leloir and Mufioz, which in our hands did not yield satisfactory oxidation of the higher fatty acids. In this paper the term cooxidant will be used to refer to the second substrate (such as fumarate, a-ketoglutarate, etc.) which must be oxidized in the presence of adenylic acid to initiate or support the oxidation of the true substrate; namely, the fatby acid.
