Abstract
ALA synthetase from R. spheroides 2 2 Abbreviations: R, Rhodopseudomonas; ALA, aminolevulinate. was purified by ammonium sulfate fractionation (0–40% saturation), Sephadex G-200 gel-filtration, and DEAE-Sephadex column chromatography. The purification study has revealed the occurrence of two types of ALA synthetase, Fraction I and Fraction II, in this bacterium. Fraction I and Fraction II were not separable from each other on Sephadex G-200 column chromatography, so that the difference in molecular weight between these two enzymes may be very slight, if any. Furthermore, one of the two types of ALA synthetase (Fraction I) was found to exist in two forms, active and inactive, in crude extracts. The inactive form of the enzyme could be converted to active form during overnight dialysis against an appropriate buffer. pH optima and K m values for substrates of the two types of ALA synthetase were compared.
