Abstract

An investigation has been made of the maximum sample loads which may be introduced onto specified gradients before the onset of droplet sedimentation in the zonal ultracentrifuge. The results showed that for the system bovine serum albumin and sucrose in the optimal reverse-wedge sample zone configuration, approximately 35% of the maximum load predicted by a simple theoretical model was obtained. Comparisons are drawn with other studies, and a more complex model is discussed in relation to practical application in zonal ultracentrifugation.
