Abstract

The ovarian ascorbic acid depletion method was used to compare the sensitivity to luteinizing hormone (LH) of the right ovary to that of the left ovary 48 hr later in the same rat. The administration of high dose levels of LH and the removal of the right ovary for assay on day 7 usually did not impair the repletion of ascorbic acid in the left ovary by day 9. The response of the left ovary to LH on day 9, however, was impaired by the previous administration on day 7 of dose levels of HCG or bovine LH that produced maximal ascorbic acid depletion in the right ovary. High dose levels of LH on day 7 reduced the responsiveness of the second ovary to the same, as well as to different, LH preparations on day 9. These findings may help explain the reduced precision of LH assays on the "second" ovary reported by some workers. It was concluded that the animals should not be used for a second assay after 48 hr unless care is taken to discard the animals that showed maximal ovarian ascorbic acid depletion on day 7.
