Abstract

Observations on the mechanism of multinucleation in primary monolayer cultures of neonatal rat muscle and rat rhabdomyosarcomas are reported. Mononuclear cells were often seen aligned with multinuclear cells in the fashion characteristic of fusing myoblasts. Mitotic figures were scattered throughout the monolayer. A vast majority of the dividing nuclei were found in mono-nuclear cells but occasionally mitoses were seen within the strap cells in both types of culture. Evidence is presented to support the hypothesis that although multinucleation of myotube-like cells in both muscle and rhabdomyo-sarcoma cultures is mainly due to fusion of cells it can also be accomplished by mitosis within the myotube.
