Abstract

Objective To establish orthotopic xenograft model of human glioblastoma multiforme (GBM) in the brain of nude mice with green fluorescent protein (GFP) expression. Methods Nude mice with ubiquitous GFP expression were obtained by crossing BALB/c nu/nu nude mice with transgenic C57/B6-GFP mice. Then, the fresh tumor tissues of human GBM were orthotopically implanted into the brains of GFP nude mice. The biologic features with growth pattern, angiogenesis and epidermal growth factor receptor (EGFR) expression were determined by fluorescence microscopy. Results GFP nude mice were generated by reciprocally crossing female C57/B6-GFP mice with male BALB/c nu/nu nude mice. The orthotopic xenografts showed invasive behavior, high EGFR expression profile with (78.6 ± 5.2 )% positive rate, and rich angiogenesis under fluorescence background. Conclusion The orthotopic xenografts from human GBM grown in the GFP nude mice shared the same biological features with high EGFR expression and invasiveness with its counterpart in the original tumors. Key words: Green fluorescent protein nude mouse; Glioblastoma multiforme; Orthotopic xenograft; Fluorescence imaging
