Abstract
Non-specific esterase (E.C.3.1.1.1) polymorphism in reference pathotypes of potato cyst nematodes (PCN) was studied by isoelectric focusing (IEF) and used to compare pathotyped field populations of PCN. Agarose IEF, pH 3-10, produced the best separation of discriminatory isoenzymes, and a distinct difference was detected between the two species of PCN with G. rostochiensis possessing an cxtra band. Six field populations of G. rostochiensis Ro1 and one population of G. pallida also possessed this band, but the band was absent from 21 other populations of G. pallida with varying pathotype composition. The usefulness of this character in the rapid identification of field populations of PCN is discussed.
