Abstract

The toxic effect of nimesulide on kidney kidney of prepubertal rats were studied on albino rats of four to five weeks old weighing between seventy to eighty grams. Different doses of nimesulide were given to the rats orally for fifty six days. The rats were divided into three groups of five rats each. The rats in the first group received no drug and acted as control. The rats in the second group received 5mg/kg/day nimesulide while those in the third group received 7.5mg/kg/day nimesulide. At the end of study, various toxicological indices were tested. The mean kidney absolute and relative weights of both treated groups were not significantly different (p>0.05) when compared with control Treatment Groups Initial Weight(g) Final Weight (g) Weight Gain(g) %Weight Gain Control 75.4±3,6 198.9± 28.6 123.5±13.2 163.2±15.8 5mg/kg/day 81.2±6.6 2007.7±13.5 119.7±8.9 150±16.0 7.5mg/kg/day 83.8±7.7 225.6±23 143.6±7.9* 174±5.2* . The serum urea and creatinine concentrations in the two treated groups were significantly increased (p蠄 0.05) when compared with control Treatment Groups Urea mg/dl Creatinine mg/kg Urea/Creatinine Ratio Control 20.3±0.2 0.64V1.4 31.89±0.6 5mg/kg/daynimesulide 29.08±0.14* 0.73±1.2* 37.81±0.4* 7.5mg/kg/daynimesulide 28.6±0.20* 0.73±1.3* 37.19±1.9* . The histopathology result showed that the rats in the low dose (5mg/kg/day) category had mild to moderate tubular necrosis, while those in the high dose group (7.5mg/kg/day) presented with moderate to severe tubular necrosis, glomerular inflammation and interstitial nephritis.
