Abstract

Background: There are no proven effective therapies for idiopathic pulmonary fibrosis (IPF) which has a 5-year survival from diagnosis of 30-50%. Rapamycin derivative (RAD) is a new macrolide immunosuppressive agent with antifibroproliferative capacity, which has been shown to inhibit platelet-derived growth factor and basic fibroblast growth factor stimulated proliferation of human adult lung fibroblasts. RAD also inhibits smooth muscle cell proliferation in the rodent heterotopic tracheal model of obliterative bronchiolitis (OB) and is under trial for the prevention of OB after lung transplantation.
