Abstract

A rapid, sensitive assay for determining ATP-sulphurylase activity in plant tissues using the firefly luciferin-luciferase system and a liquid scintillation spectrometer is described. By including pyrophosphate in the assay, it was possible to monitor the light emission from ATP which was produced from APS in this reaction. Methods have been developed that overcome interference from ATPase in both extracts of the firefly and plant tissues and that also take into account the small ATP-sulphurylase activity of firefly extracts. A computer program developed to analyze these results is presented and discussed.
