Abstract

Summary Degradation of Met-enkephalin (Try-Gly-Gly-Phe-Met) by clevage of the Tyr-Gly bond is enhanced in rodent plasma during a single or multiple dose regimen of chlorpromazine (CPZ). This is observed after five hours from time of last dose and persists as long as twenty hours later. Enhanced plasma protease activity was also observed with fluphenazine but not with imipramine. In contrast, levels of membrane-bound aminopeptidase activity in corpus striatum did not differ significantly after three weeks treatment with CPZ. In Vitro additions of CPZ (up to 100μM) to the plasma and brain preparations did not alter enzyme activity.
