Abstract
Abstract summary In this study poly(lactic-co-glycolic acid) (PLGA) nanospheres were investigated to serve as a vascular endothelial growth factor (VEGF) gene carrier to ischemic limb. Loading of VEGF gene into PLGA nanospheres sustained the release of the gene maintaining its structural and functional integrity and showing low toxicity. In mouse ischemic limbs, the PLGA nanosphere system showed significantly higher VEGF expression and more extensive neovascularization compared to naked pDNA and polyethylenimine (PEI). These results suggest that PLGA nanospheres might be useful as gene carrier for skeletal muscle gene delivery applications.
